Is there a way I can get wireshark to re-read my <tcp>.mate file?
I have been quitting and restarting wireshark to get it to read any changes to <tcp>.mate file.

No, you have to specify the path to each file.

One warning: my experience has been that the MATE documentation on the 
Wiki is quite out of date.
