I do not have personal experience fuzzing IPv6 but it seems to me like
Scapy6 [1] can help.

Something like the below might do the job:
Welcome to Scapy (1.2.0.2)
IPv6 enabled
> > > sendp((Ether()/fuzz(IPv6(dst='::1'))),loop=1)
